Animal protecing housings and animal protecting handlings gives more power to the immuno system of our rabbits with the result of much better antibodies with low backgrounds.

We use SPF New Zealand White rabbits. Housings are at 16 to 24 Celsius and 40 to 60% humidity and the filtered fresh air exchange rate is 20 times each hour.

Rabbit Antisera

Normal Rabbit antisera package. Cat.A053.
Preimmune serum, immune serum 35-80 ml from each rabbit, elisa titre detection of antiserum after the third immunizaton. If the titre is not high we do two more boosts. More boosts are frequently needed for peptide antibodies. Immunizations will start within 1-5 days after arrival of antigen.

SuperFast Rabbit Antiserum package. Cat.A055.
Comparing the titres of our SuperFast procedure for more then 12 years with normal  immunizations we can now offer this fast service to all of our customers. We use a special antigen preparation method to get a fast antigen response. The serum collection on day 28 after start of immunizations without an intermediate Elisa to test the titre. No additional boosts are possible with this service.

 

PROTEINS:
1. Amount needed:    0,2 to 0,6 mg for each rabbit is in most cases enough. In some cases it is also enough to immunize with 3 times 5 µg of antigen only. If you have only 20 to 50 µg this in in many cases enough to get good antibodies.
2. Solution: In PBS or 0,9% NaCl is perfect. The pH is best between pH  6 and pH 8. The preparation can contain 1 to 2 M Urea. Please avoid: methanol, guanidine-HCl, toxic substances, organic soluants.
3. If your protein is toxic or only soluble in organic substances we can introduce it into liposomes to overcome problems during the immunization period.
4. Concentration: 0,4 mg/ml to 2 mg/ml is best. Higher concentration are also ok. Lower concentrations are better concentrated. We offer a concentration service for only EUR 34 (Cat.No.A704)  for each antigen.
5. If your protein is within an acrylamid gel matrix this is ok. We offer the preparation of the protein in a gel matrix for only EUR 34 (Cat.A703).

 

PEPTIDES:
1. Amount needed: 1-2 mg for each rabbit or hen  for the immunizations. 3-5mg for an affinity matrix. 1 mg for Elisa tests.
2. Conjugation to KLH or BSA or Ovalbumin or Thyroglobulin or Dextran. If not otherwise indicated we use hemocyanin from moluks (KLH) as this is in most cases the best. Please make use of our peptide synthesis/ peptide conjugation combi pack to save money and time Cat.P101).
3. We can conjugate peptides via amino groups or hydroxyl groups or sulphhydril groups or carboxyl groups or other reactions on demand.

 

PHOSPHO SPECIFIC ANTIBODIES
We produce phosphospecific antibodies since 1998.
Our package includes all to get the phosphospecific antibodies you need.
a) Free peptide search with our prediction program using up to 10 algorithms.
b) Synthesis of phosphopeptide+conjugation to a carrier with 1/3 of the peptide.
c) Synthesis of non-phosphopeptide
d) Rabbit antiserum production. Preimmune, Immune (35-80 ml each rabbit),
e) Elisa with immune serum.
f) Affinity purification with phosphopeptide.
g) Affinity depletion with non-phosphopepide to remove
anti-non-phosphopeptide reaction of the antibody fraction.
h) Protocol together with the antibodies.
i)  You will get one fraction with anti-phosphospecific antibodies
